If you are a resident in Spain and you have lost your driving licence from the UK, or it has been stolen, You need to get a Certificate of Entitlement from the DVLA (or relevant authority from the country that issued the licence in other countries than UK) and apply for a new driving permit in Spain, with DGT/Trafico.
If you had previously registered your licence with Trafico, you do not need to get this certificate first, as your data are already in the Trafico database.
You do need to denounce the loss/robbery with the police and bring the denuncia with you to Trafico.
